چکیده
Objective: To evaluate the validity of the Enzyme-linked Immuno-electro Transfer Blot (EITB) technique to diagnose human hydatidosis using sheep hydatid fluid antigens and human sera infected with hydatidosis. Design: After preparing parasite antigen from sheep hydatid cyst fluid, all collected human sera infected with hydatidosis and other parasitic diseases as well as normal individuals, were analyzed by EITB test to evaluate its validity in diagnosing of hydatidosis. Setting: Department of Medical Parasitology and Mycology, School of Public Health, Tehran University of Medical Sciences, Iran. Subjects: Seventy patients infected with hydatidosis confirmed by surgery, 15 with fasciolosis, 10 with toxocariasis, 15 with strongyloidiasis, 5 with amoebiasis, 10 with trichostrongylosis and 30 normal controls. Main Outcome Measures: The sensitivity, specificity, positive and negative predictive values of EITB test. Result: Using total IgG antibody isotype, the sensitivity, specificity, positive & negative predictive values were 95%, 88.5%, 86.4%, 95.5 % 97.3%, 64.8%, 60.3% & 97.7% for antigens B&5 respectively. The total IgG antibodies in hydatidosis patients documented the parasite AgB subunits i.e 12, 16, 23 kDa, also larger subunit of Ag5, namely 39 kDa. Conclusion: The study showed that although, EITB method was a time consuming test, but due to high validity could be considered as an authentic technique, especially when the diagnosis is vague and time is not imperative. Serological Evaluation of EgAgB16 kDa, a Recombinant Antigen from Echinococcus granulosus for Diagnosis of Human Hydatidosis
BACKGROUND Regarding that accurate diagnosis of human hydatidosis still needs more investigations, the present study was conducted to clone, express, and evaluate the gene encoding AgB subunits (EgAgB16 kDa) from Echinococcus granulosus (Iranian G1 strain) and its evaluation by ELISA test. METHODS DNA was extracted from protoscoleces and was utilized by PCR for strain identification. Total RN...
متن کاملA serological survey for hydatidosis among buffaloes in Orumia
Hydatid cyst is an important and zoonotic infection caused by cystic stage of Echinococcus granulosus. It is a major and economic problem in most areas of the world that livestock are kept such as Iran. In this study, a total of 111 buffalo,s sera from Center of Buffalo Sperm Preparation (CBSP), Orumia (West Azarbayjan province, Iran, 2003) were examined for the presence of hydatidosis by usin...

Background and Objective: Hydatid cyst is one of the most common parasitic diseases of humans and livestock, which is caused by the larval stages of the Echinococcus granulosus parasite. The disease is present in all parts of the world, including Iran, and it is important for health and economics. Therefore, the aim of this study was to investigate the prevalence of hydatid cyst in the slaughte...
متن کاملSpecific recognition of hydatid cyst antigens by serum IgG, IgE, and IgA using western blot.
Diagnosis of hydatid disease in humans relies on the detection of specific antibodies against antigens of the metacestode from Echinococcus granulosus. The specificity and sensitivity of current immunological techniques based on specific serum IgG rely on the way antigens are purified. We used Western immunoblotting to detect specific IgG, IgE, and IgA antibodies in serum from patients with hyd...
متن کاملThe role of platelet microparticles in the production of antibodies from B lymphocytes against HLA-DR antigen in vitro
Background: Platelets can activate B cells and stimulate them for the production of antibodies. Since platelet microparticles (PMPs) originate from platelets, they may have the same virtue. In the present study, the effect of PMPs was investigated on the production of human leukocyte antigen (HLA)-specific antibody from B cells in vitro. Materials and Methods: In this experimental study, HLA-D...
